All the benefits of standard MegaMix but with the extra convenience of a blue agarose loading gel,
allowing for easy visualisation and accurate small volume pipetting.
The dye incorporated into the MegaMix -BLUE does not interfere with the usefulness or the further analysis of PCR products. It does not inhibit restriction enzymes or ligases, furthermore the blue dye does not flouresce at the wavelengths used by automated DNA sequencers.
MegaMix -BLUE is also available as a double concentration.
A special MegaMix formulation for the Wave® Machine. Any chemicals found in PCR buffers (eg BSA) can interfere with the performance of the Wave® Machine.
MegaMix -W does not contain any such chemicals and gives consistant and good results without further optimisation.

Used for 'difficult' PCRs, MegaMix -GOLD is a double concentrated, Hot-Start Taq mix containing optimised Taq, 
anti-Taq polymerase monoclonal antibodies, 400µM dNTPs and 6 mM MgCl2 in a buffer with a stabiliser.
The mix only becomes active when the temperature has reached >90°C.
The chance of primers forming primer-dimers or unspecific bonds to incorrect DNA sequences is
thus minimised.
MegaMix -GOLD is also available with a high or low concentration of ROX reference dye.

Five primer pairs amplify from five different
regions of the
chromosome = G on the gel photo.
If the DNA is partially degraded or fragmented one or more PCR fragments are missing = B on the gel photo.
If the DNA is fully degraded all of the five fragments are missing and only the
positive PCR control is visible = U on the gel photo.
Microzone developed the cDNAOK! kit to screen cDNA preparations before taking them on further, often very expensive, downstream processes, enabling the removal of any unsuccessful and degraded preparations saving valuable time and money.
cDNAOK! is a multiplex PCR kit, specially designed to amplify constitutively expressed genes from different chromosomes.
Careful optimisation when making this kit means that only cDNA is amplified and not any contaminating genomic DNA,
therefore giving a true reflection of the quality of the cDNA preparation.
If the cDNA preparation is good, then all four PCR
products will be amplified = G on the gel photo.
If the preparation is weak or the cDNA is degraded
then the intensity of the bands will decrease
= B on the gel photo.
If there is no cDNA present at all, or it has
completely degraded, then
only the positive control
will amplify = U on the gel photo.

G = Good machine: - Top band just visible. Machine working correctly.
B = Bad machine: - Middle and top band too strong.
Machine cooling below the required annealing temp.
U = Ugly machine: - No top band and middle band very faint.
Machine not cooling to the required annealing temp.

Tough-Taq polymerase is expressed from a 5' deletion of the gene encoding the Thermus Aquaticus DNA polymerase, resulting in a protein very similar to the Stoffle Fragment.
It is a highly active and extremely heat stable DNA polymerase, repeated exposure to 98°c in the appropriate reaction buffer does not seem to diminish the enzyme activity.In contrast the full length enzyme will not tolerate such treatment.
Each kit contains: Tough-Taq 500 or 5000 units, Reaction Buffer Plus (10x), Reaction Buffer Minus (10x), MgCl2 (1.0M) and Enzyme dilution buffer.
Tough-Taq is available in two different activities:
Kit 1 - 25 units/µl
Kit 2 - 5 units/µl
